Post-Doctoral Position in Protein Crystallography (m/f)

A postdoctoral position is available at Proteros in Martinsried

  • The subject of the project will be the structure determination of a therapeutic target of considerable interest
  • You will handle construct design, cloning, protein expression (E.coli, Bacculo or Mammalian), Purification and set up of activity assays
  • You will conduct all crystallographic activities of protein structure determination:
  • X-ray diffraction data collection (in-house generators and synchrotron beamlines)
  • X-ray diffraction data processing, experimental phasing and MR
  • model building and refinement

The ideal candidate will meet the following requirements:

  • You have a degree in Chemistry/Biochemstry/Biology or a related field and a Ph.D. in X ray protein crystallography with excellent track record
  • strong experience in molecular biology (cloning), protein production
  • PDB-depositions and related publications
  • Ideally you have good knowledge of LINUX systems and acquaintance with current software packages employed in protein crystallography.
  • You have excellent communication skills and like to work in a multi-disciplinary team

Position:The successful candidate will work on Epigenetic drug discovery. In particular, the focus of the work will be drug target validation and mode of actions of compounds targeting epigenetic factors. She/he will set strategies for studies, implement new methods and combine them with cutting edge chemoproteomic technologies. The position requires work in a multi-disciplinary team and the ability to interact with groups across different lines, GSK sites and academic labs.
Requirements:Recent PhD in biology or biochemistry (0-2 years post-doctoral experience). A background in epigenetic and chromatin biology including familiarity and experience with concepts and broad based epigenetic technologies (i.e. Chip) will be advantageous.

We have an exciting opportunity for a biochemist/biophysicist/crystallographer to join the Structure & Biophysics group at AstraZeneca in Waltham, MA. This is a collaborative effort with the laboratory of Professor Jamie Cate at the University of California Berkeley to understand the structural basis of mechanism-based toxicity to antibiotic treatment. Success in generating chimeric ribosomes will result in novel means to assess antibacterial sensitivity using a suite of genetic, biochemical and biophysical approaches.

This position provides an opportunity to explore the gene-to-structure process in an industrial setting. You'll be exposed to a variety of aspects within the drug discovery cascade, and will interact with scientists in a cross-disciplinary environment. You'll also be expected to deliver your novel research results within AstraZeneca, and externally via scientific conferences and peer-reviewed publications.

Your major responsibilities


* Build an understanding of antibiotic selectivity for the bacterial ribosome through the design and analysis of the functional activity of chimeric ribosomes
* Assess the antibacterial sensitivity of recombinant bacterial strains containing chimeric ribosomes against a panel of known ribosomal inhibitors
* Understand the structural basis of ribosomal inhibitor selectivity by performing crystallography on chimeric ribosomes in complex with antibiotics
* Work collaboratively with scientists across various disciplines to achieve project objectives
* Externally present results at scientific conferences, and publish scientific papers in high quality peer-reviewed journals


Minimum experience

A PhD in one or more of the following areas:


* Protein/RNA biochemistry
* Biophysics, crystallography, ribosome biology, molecular microbiology


Preferred experience/requirements


* Molecular biology, expression and purification skills to independently produce high-quality RNA-protein complexes
* Experience with multiple biochemical and biophysical techniques to assess the quality, stability and function of RNA-protein complexes
* Experience in performing antibacterial sensitivity assays
* Experience in designing complex reagents for biophysical and crystallographic studies
* Crystallographic experience is highly desirable

Industrial Postdoc Position in drug discovery at AstraZeneca, Sweden

Objectives
• To enable more efficient synthetic chemistry efforts in drug discovery by determining the relative orientation of small molecule ligands to at least one drug target protein.
• To show that this can be achieved within a time-frame that is compatible with the time-lines in a drug-discovery programme.

Responsibilities
• Show strong initiative in conducting research, and bringing it to functional use within AstraZeneca and the drug discovery community.
• Interface with scientists from across many disciplines while at the same time being able to work independently.
• Strong written and oral communication skills.

Tasks and methodology
• Production of drug target protein with lanthanide binding tags rigidly attached.
• Determination of pseudo-contact shift (PCS) tensors for the lanthanide tagged proteins.
• Detection of transferred PCSs for several ligands.
• Structure calculations based on PCSs. Comparison with known crystal structures to assess quality of PCS-based structures.

Minimum experience
• A PhD in biomolecular NMR or closely related field.

• Protein NMR assignments.
• Protein-ligand interactions and binding kinetics.
• Protein production.
• Paramagnetic NMR.

About BioMed X

The BioMed X Innovation Center constitutes a novel way of collaboration at the interface between academia and industry. Young life science talents from all over the world will be working jointly on pre-clinical research topics in the fields of biomedicine, molecular biology, cell biology and chemoinformatics. Interdisciplinary project teams are collaborating in an open-innovation lab facility on the campus of the University of Heidelberg, with guidance of experienced mentors while expanding their scientific network and receiving an intensive entrepreneurship and leadership training.


What we are looking for

Applications are invited from outstanding early-career scientists working or interested in one of the following fields of research:


1. Cancer & Inflammation:

The immunosuppressive microenvironment of tumors applying methods for the identification of new targets for immunotherapy of cancer

Cross-talk between signaling and metabolism in cancer and identification of new metabolic targets for cancer therapy
3. Selective Kinase Inhibitors:

What we offer

Six postgraduate research associate positions are offered for an initial term of two years, with the possibility of prolongation for up to two additional years, and an option for a permanent position. The positions include generous funding packages covering personal salary and consumables, as well as access to the core facilities of neighboring research institutions and biotechnology companies.
